1.NINHYDRIN TEST:
Common colour test for presence of aminoacid is Ninhydrin test where here aminoacid react with ninhydrin reagents to form rheumans complex .Thus ninhydrin test is test for aminoacids.
2.BIURETE TEST:
Proteins has peptide linkage can react with biurete reagent to form purple coloured complex.
3.XANTHOPROTEIC TEST:
Aromatic aminoacid like tyrosine, tryptophan and phenylalanine produces a yellow coloured complex on reaction with nitric acid.
4.MILLONS TEST:
Hydroxy aromatic aminoacid like tyrosine react with mercuric nitrate form red coloured complex.
5.HOPKINES COLE TEST:
In this test indole group containing aminoacids like tryptophan react with concentrated sulphuric acid and glyoxylic acid form violet ring .
6.EHRLISCH TEST :
Indole group consisting aminoacid like tryptophan react with para dimethyl amino benzaldehyde and 12 n hydrochloride acid form blue coloured complex.
7.PAULYS TEST:
Bulky aminoacid like tyrosine, tryptophan and histidine react with diazonium salt ( sulphanalic acid and hydrochloric acid ) form red coloured complex.
8.SAGAKUCHI TEST:
Basic aminoacid like arginine and lysin react with Alpha napthol to firm red coloured complex.
9.SODIUM NITROPRUSSIDE TEST :
Sulphur containing aminoacid like cystine and methionine with sodium NITROPRUSSIDE form red colour complex.
10.LEAD ACETATE TEST :
Aminoacid like Sulphur containing aminoacid like cystine and methionine form black precpitate with lead acetate .
